Article Snippet: The mTEC clones were incubated with 100 nmol/l of trichostatin A (A.G. Scientific) for 24 h. After the incubation, rat anti-mouse lymphotoxin -receptor antibody (IgG2a azide-free antibody; Serotec) was added to the cells at a concentration of 1 g/ml for 6 h. Real-time PCR was performed on 200 ng of cDNA, generated using random primers, both for Aire and insulin gene expression, and normalized to 18s rRNA gene expression using TaqMan Gene Expression assay kits for all genes (Applied Biosystems).
